FOXO4-dri mastery course
Unit 9 of 11

Dosing and Administration

What the community reports and why the data is extremely limited.

documenting community practice

The content in this unit documents community-reported protocols for educational purposes only. None of this constitutes medical advice. no dosing regimen described has been validated by clinical trials for the use cases discussed. product quality from unregulated sources varies and is a significant risk factor. FOXO4-DRI is among the most expensive research peptides available and human use data is extremely sparse.

dosing context at a glance

Key numbers from mouse studies and community reports.

5 mg/kg
mouse dose used in Baar et al. 2017 -- intraperitoneal or subcutaneous, 3-day cycle
1-3 mg
most commonly reported community dose per injection -- far below allometric projections
unknown
human clinical data -- zero human trials have been conducted for FOXO4-DRI
3-dose cycle
speculative community observation: some researchers note a cycle of one injection every other day for 3 doses, though this is entirely unvalidated in humans
$200-500+
approximate cost per cycle from unregulated sources -- one of the most expensive research peptides

No human dose-finding study exists for FOXO4-DRI. Community doses are rough allometric extrapolations from a single mouse study (Baar et al. 2017). Product quality from unregulated sources varies significantly and is a major risk factor.

key terms

Definitions for this unit.

A allometric scaling pharmacokinetics
A method for estimating human-equivalent doses from animal data using body surface area ratios rather than simple weight conversion. For FOXO4-DRI, allometric scaling from the 5 mg/kg mouse dose suggests roughly 0.8-1.5 mg/kg in humans -- but community doses are far lower.
S subcutaneous injection route
Injection into the fat layer between skin and muscle. The most common community-reported route for FOXO4-DRI. Slower absorption than intravenous but more practical for self-administration. The Baar et al. study used both intraperitoneal and subcutaneous routes in mice.
R reconstitution preparation
Dissolving lyophilized (freeze-dried) peptide powder in bacteriostatic water to create an injectable solution. FOXO4-DRI typically arrives as a lyophilized powder in 5mg or 10mg vials. The concentration depends on how much water is added.
C cycling protocol administration
The practice of using a peptide for a set period then taking a break. For FOXO4-DRI, community protocols mirror the Baar et al. mouse model: 3 doses over about a week, then monthly or quarterly rest periods. The rationale is that senolytic effects may be durable without continuous dosing.
B bacteriostatic water diluent
Sterile water containing 0.9% benzyl alcohol as a preservative. Used to reconstitute lyophilized peptides. The preservative allows multi-use over days without bacterial contamination, unlike plain sterile water which must be used immediately.
