BPC-157’s behavior starts with its sequence. A short, proline-rich, charge-balanced chain with no cysteines and no aromatic residues is unusually rugged for a peptide, and that ruggedness is the basis of the lab’s headline claim: that it survives stomach acid for more than a day.

BPC-157 is one short chain: Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val. Three regions do most of the explaining: the flexible glycine-studded backbone (its main chain), the stiff triple-proline run at positions 3-5, and the acidic Asp-Asp pair. That makeup matters: with four prolines, no aromatic building blocks (residues), and no cysteines, the chain has few of the soft spots digestive enzymes usually attack. It also means BPC-157 forms none of the sulfur-to-sulfur clasps called disulfide bonds, and no classical folded structure, behaving instead as a small, flexible signaling fragment.

AdvancedNo fold, no problem

Unlike larger proteins, BPC-157 has no disulfide bonds and adopts no single folded shape; it is best pictured as a flexible string whose local features, the glycine hinges and the rigid triple-proline run, do the work rather than a global structure. That flexibility is why a short fragment can still present a defined polyproline surface without needing to fold, an idea the later SH3-binding hypothesis depends on.
